You need to select an individual point on a path in order to be able to set the first vertex. To do this choose the selection tool and then click on the point on the path that you wish to be the first

If you change the layers' anchor points to the spots where you want the line to begin and end, you can use the Beam effect and pick-whip the effects start and stop coordinates to each layer's position
